Description:
Emetovoid (Tablets & Injection) is an antinauseant and antiemetic agent.

Composition:
Emetovoid TabletsEach tablet contains:
Granisetron (as hydrochloride).... 1 mgEach ampoule (1 ml) contains:
Granisetron..... 1 mg
Therapeutic indications:

The prevention of nausea and/or vomiting associated with initial and repeat courses of emetogenic cancer therapy, including high-dose cisplatin, radiotherapy in adults and children aged 2 years and above.
The prevention and treatment of postoperative nausea and vomiting in adults.
As with other antiemetics, routine prophylaxis is not recommended in patients in whom there is little expectation that nausea and/or vomiting will occur postoperatively.
In patients where nausea and/or vomiting must be avoided during the postoperative period, Emetovoid Injection is recommended even where the incidence of postoperative nausea and/or vomiting is low.
Clinical Pharmacology:

Emetovoid ampoules Granisetron is a selective 5-hydroxytryptamine3 (5-HT3) receptor antagonist with little or no affinity for other serotonin receptors, including 5-HT1; 5-HT1A; 5-HT1B/C; 5-HT2; for alpha1-, alpha2- or beta-adrenoreceptors; for dopamine-D2; or for histamine-H1; benzodiazepine; picrotoxin or opioid receptors.
Serotonin receptors of the 5-HT3 type are located peripherally on vagal nerve terminals and centrally in the chemoreceptor trigger zone of the area postrema. During chemotherapy-induced vomiting, mucosal enterochromaffin cells release serotonin, which stimulates 5-HT3 receptors. This evokes vagal afferent discharge and may induce vomiting.
In most human studies, granisetron has had little effect on blood pressure, heart rate or ECG. No evidence of an effect on plasma prolactin or aldosterone concentrations has been found in other studies.
Contraindications:

Hypersensitivity to the active substance or to any of the excipients (sodium chloride-water for injections)

Special warnings and precautions:

Granisetron may reduce lower bowel motility
ECG changes including QT interval prolongation.
Caution in patienta with cardiac co-morbidities
On cardio toxic chemotherapy
Cross sentivity between 5HT3 antagonists
Interactions with other medicinal pproductsand other forms of interaction:

Drug-frug interactions with Phenobarbital and medicinal products know to prolong QT intercal and/or which are arrhythmogenic
NO drug-drug interaction with benzodazepines, neurolwptics,antiulcer products or emetogenic cancer chemotherapies
Undesirable side effects:

The most frequent are headache and constipation which may transit
Pregnancy & laction:
There is limited data,it is preferable to avoid the use of Granisteron during pregnancy & lactation

Dose & Adminstration:
Emetovoid injection:
In prevention and treatment of acute, delayed nausea and vomoting associaed with chemo & radiotherapy:1 -3 mg (10-40mg/kg) slowly I.V. infusion up to 9 mg for adults and 3 mg for children 2 years old and above over 24 hours.
Combinasion with adrenocortcal steroid in chemotherapy: 8 - 20 mg dexamethasone before the start of cytostatic therapy or 250 mg methylprednisolone prior to the start and shortly after the end of chemotherapy
Post operative nausea and vomiting: 1mg (10mg/kg) slow I.V. up to 3 mg over 24 hours
Emetovoid Tablets:
1 mg twice daily or 2 mg once daily for up to one week following radiotherapy or chemotherapy
The first dose should be adminstrated within one hour before the start of therapy
Pediatric:
No data available.
Overdose:
In case of overdose with the injection or infusion, symptomatic ratment should be given.
Dose of up to 38.5 mg of Granisetron a single injection reported with symptoms of mild headache.

Storage:
Keep at temperature not exceeding 30°C .
